 ・ Water purification system


Elix-3(Millipore)

Do not touch the panel. Pure water (equivalent to single-distilled water) is produced automatically. Collect water by turning the knob at the bottom of the gray tank, or that of the 20 L tank above the sink. The amount of water in the gray tank is indicated on the panel.
When you collect water from the gray tank, turn the knob toward the tank against your intuition. Otherwise, the knob will be broken.


Milli-Q Jr. (Millipore)


1. Pull the tank cover towards you.
2. Remove the cap of the water tank, and fill it with the pure water collected from Elix-3.
Never fill with water other than that from Elix-3.
3. Turn on the ON/OFF switch.
4. Check the water quality on the operation panel. When the quality is good enough, turn the water collection lever toward you and collect water.
5. When you have collected water, return the lever to the original position.
6. Turn off the ON/OFF switch and keep record on the log.


 
・ High-speed cooling centrifuge CR21E (Hitachi)


1.Adjust the weights of each pair of tubes by using a balance, and place them in the symmetrical positions of the rotor.
2. Operate according to the instructions in the room.
3. Keep record on the log.
 


・ High-speed compact centrifuge CF15D2 (Hitachi)


1. For 1.5 ml (2.0 ml) tubes only.
2. Place the tubes in the symmetrical positions of the rotor.
3. Operate according to the instructions in the room.
4. Keep record on the log.
 


・ Double-beam UV spectrophotometer U-2001 (Hitachi)


1. Turn on the switch. It takes ca. 30 sec until the photometer is ready.
2. The cuvette position on your side is for specimen, on the other side for control.
3. Operate acccording to the instructions in the room.
4. Keep record on the log.
5. If you have used the cuvettes in the room, wash them according to the procedure for washing glassware, and dry them upside down in the cell holder. If you have broken them, contact the person in charge (3884).
6. Paper sheet for the printer should be set by each user.


・ pH meter(Horiba)


1. Remove the protection cap from the electrode. Wash the tip of the electrode thoroughly with DDW and keep it dipped in a beaker filled with DDW.
If you find the electrode tip dry, dip it in DDW for more than 12 hours before use.
2. Press the ON OFF key and the blue backlight is lit.
3. Calibration with standard solutions.
3a. Press F1 key. The meter is now in the calibration mode. Press F3 key to clear the previous calibration setting.
3b. Calibrate by using standard solutions (Do not dump old standard solutions into the sink but store in the tank).。
3c. During calibration, the rubber plug of the electrode should be removed.
3d. Press MEAS/ key and the measured value is indicated. When the AUTO HOLD light stops blinking, the new calibration values are set. If necessary, repeat 3b-3d.
3e. Press ESC key and the meter is now in the pH measurement mode. Now you can measure pH of your solutions.
4. Measurement. Press MEAS/ key and the pH value is indicated. AUTP HOLD blinks until the value is stabilized. During measurement, the rubber plug of the electrode should be removed.
5. When you finish measurement, press ON OFF key to turn off.
6. Wash the tip of the electrode thoroughly with DDW, wipe it and place the protection cap.。
7. Place the rubber plug.
8. When the inside of the protection cap is dry, wash the inside with DDW, remove the DDW by gentle swinging, and replenish the internal solution to wet the sponge.


・ Electronic balances (Mettler, type AG and B)

The level of each balance has been adjusted on the spot. Do not move the balance from the place where it is currently placed.

1. To turn on, press the On/Off key briefly.
2. To turn off, press the On/Off key for a while.
3. To tare, press the →O/T← key briefly.
4. To weigh, gently place the sample on the disc and wait until the round symbol on the display goes off.
5. After use, Clean any spilled chemicals with a blower brush.


・ Stereo microscopes SZ4045 and SZX-12 (Olympus)

There are two SZ4045 and one SZX-12 stereo microscopes: Of the two SZ4045's, one is with a coaxial illuminator, and the other with a transmitted light illuminator base. A dark-field illumination attachment SZ-ADD can be attached to the illuminator base. It is in the drawer of the cabinet to the left of the table on which microscopes are placed. After use, remove the SZ-ADD and place it back in the drawer. The SZX-12 is equipped with a polarizer/analyser unit.

・ Protein purification system AKTA-FPLC (Amersham Pharmacia)

1. Users of this system must be registered. Those who want to use this system should contact the person in charge.
2. To be qualified, the user should have experience of using the same or similar systems.
3. The system is in the chromatography chamber at 5 deg.
4. There will be no columns for public use. Users are expected to bring their own columns (Pharmacia prepacked columns are recommended). Visiting users may keep their columns in the chamber as long as their proposal is valid.
5. Regulations for use of this system is to be established separately.

・ Ultrafast homogenizer type K(Polytron)

*This homogenizer has been moved to another facility, because this has never been used in this room. Those who wish to use this homogenizer should contact the person in charge in advence.


1. This can be used for homogenizing various tissues, etc.
2. Mount an appropriate generator shaft.
3. Make sure that the main Sw and the power control are off, and plug the power code.
4. Place the generator shaft in the sample.
5. Turn the power control to 0 and turn on the main SW. turn the pwer control slowly from 0 to process the sample. Process the sample within 10 min or the motor will be overheated.
6. After the processing is over, turn the power control to 0 and turn off the main SW.
7. Remove the generator shaft and wash it thoroughly using detergent.

Block heater

1. The temperature can be set by pressing the (*) button and upward/downward triangle button at the same time.
2. The true temperature of the block may be different from that on display.
3. Carrying this heater to other places while its block remains inserted may cause a damage to the heater.

・ Laboratory Cart


1. The laboratory cart is to carry specimens and other objects between the room and the experimental hall. Promptly return it to the room after use.
2. Do not leave it in places other than the room.
3. Do not put on objects heavier than 10 kg.
