ABSTRACT
BL41XU is a high-flux biological macromolecular crystallography beamline. Experimental environment has been developed so that it can deal with the following targets by taking advantage of the brilliant undulator beam.:
- Data collection from small (∼10 μm) crystals
- Data collection from large unit cell (∼600 Å) crystalsii.
- Ultra-high resolution (<0.8Å) data collection using high energy X-rays (>20 keV)
- MAD (Multi-wavelength anomalous dispersion) or SAD (Single wavelength anomalous dispersion) by making use of the wide X-ray energy range, 6.5 ∼ 35 KeV
In the experimental station, a diffractometer equipped with air-bearing goniometer is installed. Two types of detector, the X-ray CCD detector (Rayonix MX225HE) and the imaging plate detector (RIGAKU R-AXIS V), can be used for diffraction data collection. Installation of cryo-cooler, CRYOCOOL-G2b-LT2 (CRYO INDUSTRIES), enables sample cooling by using either nitrogen gas or helium gas. Automatic sample mounting using sample changer SPACE is available, which enable efficient sample screening.

SOURCE AND OPTICS
This beamline employs the SPring-8 standard undulator as a light source, and a Si double crystal monochromator equipped with liquid nitrogen cooling is used to monochromatize undulator beam. The available energy range is 6.5 ∼ 17.5 keV when the first harmonics of undulator is used, and 19 ∼ 35 keV in the case of the third harmonics. The KB mirrors optics is used for beam focusing and for elimination of the higher harmonics of the undulator beam. This optics provides extremely high intensity X-rays with a size of 80 x 40μm2 at the sample position. The X-ray is further collimated by a pin hole just placed before the sample, and beam size of 10, 30 and 50 μm is currently available.
